KE30001 is a qualitative measure of human anti-SARS-CoV-2 N IgG protein in plasma. The principle of the kit is indirect ELISA. The protein N recombinant protein has been pre-coated in the well of the microplate. Samples or standards are added to the well, after incubation the wells are washed and a horseradish peroxidase-conjugated anti-human IgG is added to each well.
Production of a “recombinant protein-anti-NP human IgG antibody-HRP-conjugated antibody” complex. after incubation, the wells are washed, followed by a tetramethylbenzidine (TMB) reagent. A solution containing sulfuric acid is used to stop color development and the intensity of the color, which is proportional to the amount of bound protein, can be measured at 450 nm with the correction wavelength set at 630 nm.

Coronaviruses are enveloped viruses with a positive-sense RNA genome and a helically symmetric nucleocapsid. Coronavirus nucleoproteins are located in the cytoplasm and nucleolus, a subnuclear structure, both in virus-infected primary cells and in cells transfected with plasmids that express protein N. Coronavirus N protein is necessary for the synthesis of coronavirus RNA and it has RNA chaperone activity that may be involved in template switching.
The nucleocapsid protein is one of the most abundant proteins in the coronavirus. During virion assembly, protein N binds to viral RNA and leads to the formation of the helical nucleocapsid. The nucleocapsid protein is a highly immunogenic phosphoprotein also involved in the replication of the viral genome and in the modulation of cell signaling pathways. Due to the conservation of the N protein sequence and its strong immunogenicity, the coronavirus N protein is chosen as a diagnostic tool.
COVID-19 antibodies can be produced by the host’s immune system after exposure to SARS-CoV-2. IgG and IgM antibodies are also known as IgG and IgM immunoglobulins, respectively, and are among the isotypes of antibodies produced by vertebrate immune systems. The ELISA microplate is coated with the SARS-CoV-2 nucleocapsid protein (N). The coated protein N binds to COVID-19 IgG N antibodies in the serum sample.
